| 摘 要： | 目的研究重楼总皂苷对人永生化角质形成细胞株(HaCaT细胞)增殖和分泌白介素8(IL-8)的影响。方法采用MTT法进行细胞增殖实验,采用ELISA法测定重楼总皂苷对HaCaT细胞分泌IL-8的情况。结果在所选浓度范围内(60μg/mL,90μg/mL,120μg/mL和150μg/mL),重楼总皂苷对HaCaT细胞的增殖有抑制作用;其中120μg/mL和150μg/mL浓度组重楼总皂苷可抑制HaCaT细胞自分泌IL-8,90μg/mL,120μg/mL和150μg/mL浓度组重楼总皂苷能抑制TNF-α(浓度为100U/mL),刺激HaCaT细胞分泌IL-8。结论重楼总皂苷能抑制HaCaT细胞增殖和IL-8的分泌。

| Abstract: | Objective To study the effect of steroidal saponins from P.polyphylla on proliferation of HaCaT cells and secretion of IL-8.Methods The inhibition rate of steroidal saponins from P.polyphylla for proliferation of HaCaT cells was detected by MTT.The influence of steroidal saponins from P.polyphylla on secretion of IL-8 was determined by ELISA.Results Within the selected concentration(60μg/mL,90μg/mL,120μg/ mL,150μg/mL),steroidal saponins from P.polyphylla inhibited the proliferation of HaCaT cells compared with control groups.The spontaneous secretion of IL-8 from HaCaT cells was decreased after treatment with steroidal saponins(120μg/mL,150μg/mL),meanwhile,The secretion of IL-8 from HaCaT cells by TNF-αstimulation was inhibited after treatment with steroidal saponins(90μg/mL,120μg/mL,150μg/mL). Conclusion Steroidal saponins from P.polyphylla can suppress the growth of HaCaT cells and inhibite secretion of IL-8 from HaCaT cells. |
