异丙酚对离体气管纤毛摆动频率的影响

摘    要:目的 观察异丙酚对离体气管上皮细胞纤毛摆动频率(CBF)的影响。方法 从8例喉癌病人气管造口手术中获取离体气管标本,每例标本分离出4块纤毛摆动活跃的组织块。生理盐水稀释异丙酚,终浓度分别为0.01、1、10、20μg·ml~(-1),组织块依上述浓度分为4组,每块标本随机加入一种浓度异丙酚,在加入前及加入后5、10和20min,使用相差显微镜及成像分析技术测量纤毛上皮的CBF。结果 0.01μg·ml~(-1)组及1μg·ml~(-1)组,CBF在各时间点与基础值比较差异均无显著性(P>0.05);10μg·ml~(-1)组和20μg·ml~(-1)组,CBF在各时间点与基础值比较,差异有显著性(P<0.05)。各组间的CBF基础值差异不显著(P>0.05);在其他时间点比较,异丙酚浓度增加,CBF也增加,组间差异有显著性(P<0.05)。结论 异丙酚增加离体气管上皮细胞纤毛摆动频率。

Abstract:Objective To investigate the effects of different concentrations of propofol on cilia beat frequency (CBF) of isolated human tracheal epithelium. Methods The trachea specimen was obtained from eight patients undergoing radical operation for larynx cancer. The mucous membrane of each specimen was cut into 4 small pieces under stereoscope and active beating of cilia was observed . Four concentrations of propofol in normal saline were prepared (0.01, 1, 10, 20ug.ml-1) . The 4 small pieces of mucous membrane from each patient were randomly dipped into the propofol solutions of 4 different concentrations as described.CBF were measured using phase-contrast microscope and videotape analysis before (T0, baseline) and 5 (T1 ), 10 (T2), 20 (T3) min after the specimen was dipped into propofol solution. Results Propofol 0.01 ug.ml-1 and 1 ug.ml-1 did not significantly change CBF, but propofol 10 ug.ml-1 and 20 ug.ml-1 significantly increased CBF at T1,2,3 as compared with the baseline, in a dose-dependent manner (P < 0.05) .Conclusion Propofol increases CBF of human tracheal epithelium.
