| 摘 要： | 目的:研究注射用左亚叶酸钙在健康志愿者体内的药动学。方法:采用随机、开放、平行实验设计,将30名志愿者分成A、B、C3组,每组10人,静脉滴注高、中、低3个剂量的左亚叶酸钙后,用HPLC法测定各时间点血中主药的浓度,计算药动学参数。结果:静脉滴注(60min内)高(300mg)、中(200mg)、低(100mg)剂量的左亚叶酸钙后,主要药动学参数分别为:cmax(22.05±3.28)、(13.85±4.12)、(6.99±1.08)μg/mL;tmax(1.00±0.12)、(0.98±0.28)、(1.03±0.08)h;t1/2(0.68±0.1),(0.72±0.17)、(0.64±0.12)h;AUC0→∞(28.61±3.13)、(18.23±5.47)、(9.52±1.70)μg/mL。t1/2、Vd和MRT在不同剂量组间有显著性差异(P<0.05),cmax、AUC0→3h和AUC0→∞经剂量校正后进行方差分析,与给药剂量呈良好线性关系(P<0.05)。结论:静脉滴注左亚叶酸钙的药动学符合一室模型。
