慢性丙型肝炎患者HCV-RNA含量与CD4^＋CD25^＋调节性T细胞的相关性研究

目的探讨慢性丙型肝炎（CHC）患者外周血中CD4＋CD25＋调节性T细胞含量与HCV-RNA的相关性。方法用流式细胞仪检测80例CHC患者和30例健康对照者外周血CD4＋CD25＋Treg细胞表达,荧光定量PCR检测HCV-RNA含量。结果 CHC患者外周血中CD4＋CD25＋Treg细胞明显高于健康对照组（P〈0.01）,且随HCV-RNA载量增高,患者外周血中CD4＋CD25＋Treg细胞的含量逐渐升高（P〈0.01）。CD4＋CD25＋Treg细胞与HCV-RNA取对数后呈正相关（r=0.588,P〈0.01）,与ALT呈正相关（r=0.306,P〈0.01）。结论 CHC患者外周血中CD4＋CD25＋Treg细胞增高,且与HCV-RNA含量及ALT升高具有一致性。

Study on relationship between CD4+,CD25+ regulatory T cells and HCV-RNA load in peripheral blood of patients with chronic hepatitis C

Objective To explore the amount of CD4 ^＋ , CD25 ^＋ regulatory T cells in peripheral blood of patients with chronic hepatitis C （ CHC ） and to study the relationship between these cells and HCV - RNA load. Methods The amount of CD4 ^＋ , CD25 ^＋ regulatory T cells in peripheral blood of 80 patients with chronic hepatitis C （CHC） and 30 healthy controls were detected by flow cytometry. The load of HCV - RNA was detected by fluorescent quantitative polymerase chain reaction （ FQ - PCR） assay. Results The amount of CD4 ^＋ , CD25 ^＋ regulatory T cells in peripheral blood of patients with chronic hepatitis C was significantly higher than those of control group （ P 〈0.01 ）, and the amount of CD4 ^＋ , CD25 ^＋ regulatory T cells in peripheral blood of patients was gradually increased following the increase of HCV - RNA load （ P 〈 0.01 ）. There was a positive correlation between amount of CD4 ^＋ , CD25 ^＋ regulatory T cells and HCV - RNA load after taking logarithm （ r =0. 588, P 〈0.01 ）, and it was same as ALT （ r = 0. 306, P 〈 0.01 ）. Conclusion The increase in amount of CD4 ^＋ , CD25 ^＋ regulatory T cells in patients with CHC is in accordance with copies of HCV DNA and increased level of ALT.
