摘    要:目的建立检测人B组、C组轮状病毒(RV)的膜芯片。方法采用化学合成法合成B组、C组RV高度保守的核酸扩增区域,以地高辛标记上游引物,经半巢式PCR扩增B组、C组RV的保守检测区域,采用本实验室自行研制的膜芯片进行杂交检测。结果B组、C组RV的扩增产物同膜芯片上特异探针呈高度特异杂交。结论自制的膜芯片检测技术能够实现B组、C组RV的检测.并具有特异性好、高通量、平行性和简便快速的特点。

Abstract:Objective:To develop a method of membrane gene chip for detection of human B and C rotavirus.Methods:To synthesize the highly conservative nucleotide sequences of group B and group C rotavirus and mark the up-stream prime with DIG,the conservative region for detection were amplified by semi-nested PCR.Then the PCR products were detected by hybridizing with membrane chips respectivly.Results:The products of B,C rotavirus can hybridize with the probes immobilized on the membrane chip with high specificity.Conclusion:This type of membrane chips can be used to detect human B and C rotavirus,which is distinctive,robust,parallel,quick and simple.
