两株粉,尘螨基因组DNA的研究

摘    要:了解粉螨与尘螨基因组DNA特征,以在基因水平上探讨二者的相互关系,方法:通过限制性酶切片段多态性分析和随意扩增的多态DNA(RAPD)分析,对二种粉螨及尘螨基因组DNA进行比较。结果限制性内切酶EcorI、PstI消化腐酪食螨,屋尘螨基因组DNA存在不同的限制性酶切图谱;选用4种机引物对腐酪食螨及屋尘进行扩增,仅引物PBR322/EcorI扩增片段显示出各自特有带型,计算二者RAPD相似率为64.

Study on the Genomic DNA of T.Putrescentiae and D.Pteronyssinus
Abstract:Objective:Through the analysis of the characteristics of genomic DNAs of Tgrophagus Putrescentiae and Permatophagoides Pteronyssinus to explore the relationship between them.Method:Restriction fragment length Polymorphisms (RFLP) with E COR I and PstI and random amplified Polymorphic DNA (RAPD) with four Primers were performed for comparison.Results:The RFLP patterns of T.putrescentiae and D.pteronyssinus showed significant differences.In RAPD only the PBR322/E COR I Primer yielded Characteristic patterns and there were some differences of the share random amplified DNA fragment between T.Putrescentiae and D.Pteronyssinus.The similarity rate calculated was 64.4%.Conclusion:There are heterogeneity between T.Putrescentiae and D.Pteronyssinus that can be distinguished by gene analysis,but there also exist some similarities.
