摘    要:目的 探讨Profiler plus试剂盒在DNA数据库建设中应用的适用性以及其9个常染色体短串联重复序列(short tandem repeats,STR)基因座在标准三联体亲缘关系DNA数据库比对中认定亲权的危险性.方法 在一杀人案例中,将死者和胎儿的DNA数据在全省DNA数据库中进行亲缘关系比对获得比中记录43条,组成43个假定的标准三联体家庭.计算其父权指数(paternity index,PI)值与累计父权相对机会(relative chance of paternity,RCP)值;应用Identiflier试剂盒加做到1 5个常染色体STR基因座,观察其排除情况.结果 在43例假定三联体中PI值最低为1 724.318 71138(RCP=99.942 039 69%),最高为55 178.198 764 14(RCP=99.998 187 72%),加做到15个STR基因座后,43例假定三联体中每例至少2个基因座出现排除,最多6个基因座出现排除的现象,平均排除基因座数为3.63个.结论 Profiler plus试剂盒不适合用于DNA数据库建设;其9个STR基因座应用到亲缘关系鉴定中是危险的;单纯利用RCP值来认定亲缘关系是不安全的;建议应用16个或更多的基因座建设DNA数据库,并在亲缘关系鉴定时应用16个以上的基因座进行判定.
Abstract:
Objective To explore the applicability of Profiler plus kit in constructing DNA database and the reliability of 9 short tandem repeats from Profiler plus kit in identifying Mother-Child-Father paternity in DNA database. Methods To collect 43 random persons who are matched with known Decedent(Mother) and embryo( Child )in homicide cases, we constructed 43 fictitious trio families. paternity index ( PI ) and relative chance of paternity (RCP) were computed. We obtained 15 short tandem repeats data with Identiflier kit and studied the situation of exclusion. Results The PI in 43 fictitious trio families is from 1 724. 318 711 38 ( RCP = 99. 942 039 69% ) to 55 178. 198 764 14 ( RCP = 99. 998 187 72% ). The number of exclusion locus is from 2 to 6, the average is 3.63. Conclusions Using Profiler plus kit to construct DNA database is not suitable. Paternity testing with the 9 short tandem repeats locus is not reliable. Confirmation of affinity relationship via using the RCP is not reliable either. It is necessary to establish DNA database through 16 or more loci. We recommend using 16 loci or more in paternity testing.
