氯菧酚胺对小鼠活体精原细胞姐妹染色单体交换率的影响

本实验用BrdU(5-溴脱氧尿嘧啶)替代胸腺嘧啶核苷的体内实验方法,检验了氯菧酚胺短期灌服小白鼠后精原细胞姐妹染色单体交换(SCE)频率的改变。结果表明各服药组的SCE率与对照组无明显差异。因而从生殖细胞SCE这一敏感指标提供的证据,支持氯菧酚胺不具有明显诱变作用的观点。

STUDY OF CLOMIPHENE CITRATE EFFECTS ON SISTER CHROMATID EXCHANGES (SCE) IN MOUSE SPERMATOGONIA IN VIVO

Substitution of thymidine with 5-Brome-deoxyuridine (BrdU) was performed in vivo in mice for the detection of clomiphene citrate effects on SCE in sperniatogonia. Twenty four adult male mice were divided into 8 groups. One group served as control and the other groups were given clomiphena for 5 days in doses 10, 25, 50, 75, 100 times that of clinical dose (25 mg per day) respectively. Then, all animals were injected intraperitoneally 0.5 ml of suspension of BrdU adsorbed with active carbon and were killed 54 hours later after infcraperitoneal injection of colchicine (4mg/kg). Chromosome samples of spermatogonia were prepared routinely from the testis. The results of examination indicated that difference between administration groups and control group was not statistically significant (P>0.05).
