Abstract:Objective To study the effect of withdrawal of androgen on expression of cell cycle regulation proteins in human prostate carcinoma cells. Methods After treatment of 1×10-9mol/L for 24 h, expression of cell cycle regulation molecules including CDK2, CDK4, CDK6 and p27kipl in human prostate carcinoma cell line LNCaP and PC-3 was detected by using immunoprecipitation and Western blot analysis after 0, 8 and 24 h of withdrawal of androgen. Results Down-regulation of CDK2, CDK4 and CDK6 and up-regulation of p27kipl were found initially in LNCaP cell line after 8 h withdrawal of androgen. The levels of CDKs were decreased continuously and reached the lowest after 24 h, and the most significant decrease was detected in CDK2, while continuously increase of p27kipl was detected until 24 h. No changes of expression of CDKs and p27kipl were detected in PC-3 cell line. Conclusion Down-regulation of CDK2, CDK4 and CDK6 and up-regulation of p27kipl may be one of the mechanisms by which androgen dependent prostate carcinoma could be controlled after androgen ablation by either operative or medicine methods.
