舌鳞癌中p34cdc2与cyclin B1的表达及相关研究

目的研究舌鳞癌组织中p34cdc2与cyclin B1蛋白的表达,探讨它们阳性表达的相关性及其与舌鳞癌临床病理特征的关系。方法随机收集舌鳞癌标本60例,对照上皮包括正常舌组织11例、癌旁组织49例。采用免疫组织化学SP法显示p34cdc2与cyclin B1蛋白的表达,并结合舌鳞癌的临床病理特征进行分析。结果舌癌组织中可见p34cdc2与cyclin B1蛋白表达并显棕色/黄色,主要定位于细胞浆中。两种蛋白在癌组织中存在增多表达,与正常组织和(或)癌旁组织中的表达差异有显著性(P<0.01)。p34cdc2蛋白的表达在舌鳞癌不同临床分期、组织分级和有无淋巴结转移组间差异有显著性(P<0.05),而与年龄、性别无统计学意义;同时cyclin B1蛋白的表达也在舌鳞癌不同临床分期、组织分级中差异有显著性(P<0.05),而在不同年龄、性别及有无淋巴结转移组间差异无显著性。结论p34cdc2及cyclin B1蛋白在舌鳞癌中有表达增多现象,或许可反映舌鳞癌细胞分裂增殖能力;增多表达的p34cdc2及cyclin B1蛋白可作为评价舌鳞癌临床分期和组织分级等临床病理特征的参考指标之一。

The expression and significance of p34cdc2 and cyclin B1 in tongue squamous cell carcinoma

Objective To analyze the expression and the correlation of p34cdc2 and cyclin B1 in tongue squamous cell carcinoma(SCC),and furtherly explore the relation between the two proteins with the clinicopathological features of tongue SCC. Methods 60 cases of tongue SCC were selected randomly. The epithelia of 11 healthy tongue tissues and 49 neighboring noncancerous ones were selected as the control. These samples were detected by immunohistochemiscal method,and the clinicopathological features of tongue SCC were analyzed. Results The expression of p34cdc2 and cyclin B1 were basically localized in cytoplasm of tongue SCC cells,showing brown/yellow. We found over-expression of the two proteins in tongue SCC.The difference was statistically significant between the expression of p34cdc2 and cyclin B1 in cancer tissues and healthy or/and neighboring non-cancerous tissues (P<0.01). Expression of p34cdc2 was statistically different among clinical stages,histological grades and lymphatic metastasis of tongue SCC (P<0.05),which was the same as cyclin B1 expect of lymphatic metastasis.No statistical significance was found between different age,sex and the expression of the two protein. Conclusion Over-expression of p34cdc2 and cyclin B1 are found in tongue SCC.Overexpression of the two proteins may be one of good marks in showing the proliferative ability of tongue SCC and determining the clinical stages,histological grades and other clinicopathological features of tongue SCC.
