原位移植人肝癌裸鼠模型AFP基因甲基化状态与其表达的关系

【目的】探讨原位移植人肝癌模型肝癌组织中，AFP基因启动子区的甲基化状态同其异常表达的关系。【方法】采用甲基化特异性PCR（MSP）和亚硫酸氢盐测序法（BSP）分析模型肝癌组织中AFP基因启动子区甲基化状态；RT-PCR及Western Blotting两种方法检测AFP基因在转录水平和翻译水平的表达情况。【结果】10例肝癌模型的肝癌组织均有AFP基因mRNA和蛋白的表达（100％），AFP基因启动子区部分CG位点发生异常甲基化，其中－2431、－2494两个位点低于正常对照（P〈0．01）。【结论】AFP基因启动子区甲基化程度与其表达成负相关（P〈0．01）。

The methylation status and expression of AFP gene in orthotopic implanted human primary hepaticcarcinoma in nude mice models

[ Objective] To expiore the relation between methylation status and expression of AFP gene in orthotopic implanted human primary hepaticcarcinoma in nude mice models. [Methods] The mehtylation status of the AFP gene in the liver cancers from 10 orthotopic implanted nude mice models of human hepatocellular carcinoma were detected and analized by MSP and BSP. The expression of AFP gene were detected by RT-PCR and Western Blotting. [ Results] The expression of AFP gene in the liver cancer were in all the mice （100%） . In the promoter region of AFP gene some CG sites changed, in which two sites were lower than controls （ P 〈 0.01 ）. [ Conclusions] There is a negative relation between the methylation status and expression of AFP gene in the promoter region （ P 〈 0.01）.
