C-erbB-2、Cath-D和MMP-9在胃癌中的表达及其临床意义

目的:探讨癌基因C—erbB-2、组织蛋白酶D(Cath—D)和基质金属酶-9(MMP-9)在胃癌中的表达及其与胃癌生物学行为的关系。方法:采用免疫组织化学(S-P)法,检测了87例胃癌标本中C—erbB-2、Cath-D和MMP-9的表达。结果:87例胃癌中C—erbB-2阳性表达占47.13％(41/87);Cath—D阳性表达占53.57％(45/87);MMP-9阳性表达占80.95％(68/87);胃癌中C—erbB-2的阳性表达与胃癌浸润深度(P<0.05)、淋巴结转移(P<0.01)密切相关。胃癌中侵及浆膜层MMP-9表达率高于侵及肌层组,侵及肌层组MMP-9表达率高于浸润黏膜层及黏膜下层组(P<0.05),伴有淋巴结转移组高于不伴有淋巴结转移组,但统计学上无显著差异(P>0.05)。Cath—D阳性表达与胃癌的分化程度(P<0.05)、浸润深度(P<0.05)及淋巴结转移(P<0.01)有关。结论:C—erbB-2、Cath—D和MMP-9与胃癌的生长、浸润、转移有密切关系,可作为判断胃癌生物学行为和预后的重要指标。

Expression of C-erbB-2, Cathepsin D and Matrix Metalloproteinase-9 in Gastric Carcinoma and Their Clinical Significance

Objective To investigate the expression C-erbB-2 oncogene, Cath-D and MMP-9 in gastric carcinoma and their correlation with the biological behavior of gastric carcinoma. Methods Expression of C-erbB-2, Cath-D and MMP-9 in 87 cases of gastric carcinoma were detected by S-P immunohistochemical method. Result The positive rates of the expression of C-erbB-2. Cath-D and MMP-9 in 87 cases of gastric carcinoma were 47. 13 % . 53. 57 % and 80. 95 % respectively. The positive rates of C-erbB-2 in gastric carcinoma were correlated with depth of invasion (P<0. 05) and lymph node metastasis (P<0. 01); The positive rates of MMP-9 in gastric carcinoma invading seroral layer were higher than those invading muscular layer and undermucosal layer(P<0. 05); The positive rates of MMP-9 in gastric carcinoma with lymph node metastasis were higher than those without lymph node metastasis , but there were no signifinant in statistics; The positive rates of Cath-D in gastric carcinoma were correlated with differentiated degress, depth of invasion and lymph node metastasis (P<0. 05). Conclusion The results suggest that expression of C-erbB-2, Cath-D and MMP-9 is closely related to growth, invasion, metastasis and prognosis of gastric carcinoma.
