HPLC法测定固本咳喘胶囊中补骨脂素及异补骨脂素的含量

目的建立高效液相色谱法测定固本咳喘胶囊中补骨脂素及异补骨脂素含量的测定方法。方法色谱柱：安捷伦ZORBAX C18（4.6mm×150mm,5μm）;柱温：30℃;检测波长：246nm;流速：1.0mL/min;流动相：甲醇-0.1mol/L磷酸氢二钠溶液（用30%磷酸溶液调节pH值至7.0）（45∶55）。结果补骨脂素及异补骨脂素分别在0.02134～0.42680μg（r=0.9999,n=6）、0.02184～0.43680μg（r=0.9999,n=6）范围内,峰面积与进样量呈良好的线性关系;平均回收率分别为102.86%、105.83%;RSD值分别为1.76%（n=6）、1.08%（n=6）。结论本测定方法准确、重现性好,为固本咳喘胶囊的质量控制提供了可靠依据。

The determination of the content of psoralen and isopsoralen in Gubenkechuan Capsule by HPLC method

Objective To establish a method to test the content of psoralen and isopsoralen in Gubenkechuan Capsule by using high performance liquid chromatography. Methods The chromatography column was Agilent ZORBAX C18 （4.6 mm×150 mm, 5 μm）, the column temperature was 30℃, the detection wavelength was 246 nm, and the flow rate was 1.0 mL/min. The methanol-0.1 mol/L and disodium hydrogen phosphate solution （with 30% phosphate solution to adjust pH value to 7.0） （45∶55） was used as mobile phase. Results Psoralen and isopsoralen were in the range of 0.021 34-0.426 80 μg, r=0.999 9 （n = 6） and 0.021 84-0.436 80 μg, r=0.999 9 （n=6） respectively, there were a good linear relationship between the peak area and sample size. The average recovery rates were 102.86% and 105.83% respectively. RSD were 1.76% （n=6） and 1.08% （n = 6） respectively. Conclusion It is a accurate determination method with good reproducibility, and it can provide reliable basis for the quality control of Gubenkechuan Capsule.
