外加磁场诱导超顺磁氧化铁标记骨髓间充质干细胞迁移 的体外研究

摘    要: 【目的】 观察外加磁场能否诱导用超顺磁氧化铁标记的大鼠骨髓间充质干细胞在体外条件下定向迁移 【方法】 使用超顺磁氧化铁标记大鼠骨髓间充质干细胞,采用普鲁士蓝染色鉴定其标记率台盼蓝染色检测标记细胞生存能力,MTT法检测标记干细胞的增殖活力外加磁场进行划痕试验及Transwell试验验证磁场对标记细胞的定向诱导迁移作用 【结果】 普鲁士蓝染色显示骨髓间充质干细胞的超顺磁氧化铁标记率接近100%,台盼蓝染色标记细胞生存率与对照组无差异(P > 0.05),MTT检测标记干细胞的增殖活力与未标记干细胞无差异(P > 0.05)外加磁场可加速标记细胞填补划痕试验空白区的速度,同时可增加transwell试验跨膜的细胞数(P < 0.05),而未加磁场情况下标记细胞与未标记细胞迁移速率无差异(P > 0.05)。 【结论】 外加磁场可在体外条件下诱导超顺磁氧化铁标记的骨髓间充质干细胞定向迁移

Abstract:【Objective】 The study aims to label rat bone marrow mesenchymal stem cells with superparamagnetic iron oxide, then observe whether the magnetic field could induce labeled cells targeted migration in vitro. 【Method】 Rat bone marrow mesenchymal stem cells were labeled with superparamagnetic iron oxide and evaluated the labeling efficiency by Prussian blue staining. The viability and proliferation activity of labeled stem cells were evaluated with trypan blue staining and MTT assay respectively. Wound healing assay and transwell culture system were used to test the migration effect of external magnetic field on labeled stem cells.【 Results】 Prussian blue staining showed that mesenchymal stem cells labeled superparamagnetic iron oxide close to 100%. Trypan blue staining and MTT assay were no significant difference between labeled group and control group(P > 0.05). External magnetic field can accelerate the labeled stem cells to fill the blank areas in the wound healing and increase the number of cells crossing the membrane in the transwell culture system(P < 0.05), while without the magnetic field labeled cells and unlabeled cells were no difference in migration rate(P > 0.05). 【Conclusion】 External magnetic field can induce bone marrow mesenchymal stem cells labeled with superparamagnetic iron oxide to migrate with target in vitro.
