低幅高频振动对去卵巢大鼠植入体骨整合的影响

摘    要:【目的】 探讨低幅高频振动在促进骨质疏松大鼠骨床中植入体骨整合中的作用【方法】 共40只SD雌性大鼠随机分为4组:单纯去卵巢组(OVX),去卵巢加振动处理组(VIB),去卵巢加阿仑膦酸钠组(ALO),假手术组(Sham)。前3组行双侧去卵巢术,Sham组只暴露双侧卵巢但不予切除4周后,在所有大鼠双侧胫骨上段植入羟基磷灰石包被的圆柱形钛质植入体(长10mm,直径1mm)。然后对VIB组和ALO组分别进行机械振动(0.3g,35 Hz)和口服阿仑磷酸钠(每周7 mg/kg)处理,持续8周然后处死所有大鼠,取大鼠双侧股骨胫骨测量股骨骨密度;对一侧胫骨行假体顶出试验,计算最大顶出力和界面剪切强度;另一侧胫骨行骨组织形态学检测,测量骨整合率(BIC)和植入体周围骨量分数(BA)。【结果】 VIB组骨密度为(0.228 ± 0.022)g/cm2最大顶出力为(180 ± 22)N界面剪切强度为(6.5 ± 0.8)N/mm2BIC为(69 ± 4)%BA值为(72.7 ± 1.8)%,均显著高于OVX组分别为:(0.202 ± 0.022) g/cm2,(119 ± 16) N,(4.3 ± 0.6)N/mm2,(51 ± 5)%,(45 ± 4)%;P < 0.05],但显著低于ALO(0.254 ± 0.036) g/cm2,(224 ± 28)N,(8.3 ± 0.9) N/mm2,(83 ± 6)%,(80 ± 3)%]和Sham组(0.261 ± 0.028)/cm2,(245 ± 33)N,(8.9 ± 1.2) N/mm2,(91 ± 5)%,(89 ± 6)%;P < 0.05]。ALO组和Sham的以上各参数之间差别无统计学意义(P > 0.05)。【结论】 低幅高频机械振动可以显著促进去卵巢大鼠骨床中植入体的骨整合,但其效果要弱于阿仑膦酸钠

Abstract:【Objective】 To investigate the effect of low-magnitude high-frequency (LMHF) whole body vibration on bone-implant osseointegration in osteoporotic rats. 【Methods】 Thirty female rats were ovariectomized to induce osteoporosis, and then treated with LMHF vibration (VIB) or alendronate (ALO) or a control treatment (OVX). Another 10 rats were sham operated as Sham control group. Before treatment, hydroxyapatite (HA)-coated titanium implants were inserted into proximal tibiae bilaterally. Both LMHF vibration and alendronate treatment were lasted for 8 weeks. 【Results】 The BMD (0.228 ± 0.022) g/cm2], maximum push out force (180 ± 22) N], interfacial shear strength (6.5 ± 0.8)N/mm2], bone-implant contact BIC,(69 ± 4)%] and bone area BA,(72.7 ± 1.8)%] of group VIB were significant higher than those of group OVX (0.202 ± 0.022)g/cm2,(119 ± 16)N,(4.3 ± 0.6) N/mm2,(51 ± 5)%,(45 ± 4)%, respectively;P < 0.05], but were lower than those of group ALO(0.254 ± 0.036)g/cm2,(224 ± 28) N, (8.3 ± 0.9)N/mm2, (83 ± 6)%,(80 ± 3)%)] and Sham (0.261 ± 0.028) g/cm2,(245 ± 33)N,(8.9 ± 1.2)N/mm2,(91 ± 5)%, (89 ± 6)%;P < 0.05]. No significant difference was found between group ALO and Sham. 【Conclusion】 LMHF whole body vibration enhances bone-implant osseointegration in ovariectomized rats, but with less effectiveness when compared with alendronate.
