摘    要:目的 探讨新生大鼠神经干细胞的分离培养方法及其外源报告基因在神经干细胞中的表达情况。方法 采用无血清细胞培养技术,间接免疫细胞化学染色法及其绿色荧光蛋白转导技术。结果 从新生大鼠脑组织分离的细胞具有连续克隆能力,表达神经上皮干细胞蛋白,诱导分化后的细胞表达成熟神经细胞和星形胶质细胞特异性的蛋白。绿色荧光蛋白报告基因转导神经干细胞后能够有效表达。结论 该方法分离培养的神经干细胞保持了干细胞的未分化属性,具有自我更新、增殖能力和多分化潜能,而且外源报告基因在神经干细胞中能够有效表达。

Abstract:Objective To study the isolation and cultiv ation of neural stem cells in neonatal rats. Methods Cell culture, indirect immunofluo rescence cytochemistry and gene transfection techniques were used. Results The neural stem cells isolated in newborn rats had the potential to form clones, express neuroepithelial stem cell protein(nestin) and differentiate into mature neurons and astrocytes. Moreover, green fluorescent protein vector could be efficiently transfected into this cell line. Conclusions Neural stems cells can only be characterized on a critical functional basis in terms of their undifferentiated features, capacity of self-renewal, proliferation and pluripotentiality, and can express ex vivo gene efficiently.
