肿瘤干细胞标记物CD133与基质金属蛋白酶-9在乳腺癌组织中的表达及生物学意义

摘    要:目的探讨乳腺癌组织中基质金属蛋白酶-9(MMP-9)及肿瘤干细胞标志(CD133)的表达及其与乳腺癌临床病理参数之间的关系。方法采用免疫组织化学方法,检测109例乳腺癌组织中的CD133及MMP-9蛋白的表达并对结果进行分析。结果①109例乳腺癌组织中MMP-9、CD133蛋白的表达率分别是40.4%(44/109)、46.8%(51/109),随肿瘤直径增大以及肿瘤分期的增加,MMP-9蛋白的阳性表达率也明显增加,差别有统计学意义(P〈0.05);而乳腺癌组织中CD133+肿瘤干细胞数量则随病理级别的升高而增加(P〈0.05)。②109例乳腺癌组织中,CD133+肿瘤干细胞MMP-9的阳性表达随病理分级增加而增加(P〈0.05)。结论低分化乳腺癌组织中,MMP-9蛋白与CD133+肿瘤干细胞存在密切关系,CD133+肿瘤干细胞中MMP-9蛋白的表达可能参与乳腺癌恶性演进,或可成为预后预测的指标之一。

Expression and significance of CD133 and MMP-9 in breast cancer

Abstract:Objective To investigate the expression of matrix metalloproteinase-9 (MMP-9) and CD133 protein in breast cancer and their relationship with the clinical characteristics of breast cancer.Methods 109 cases of breast cancer tissues by immunohistochemistry for MMP-9 and CD133.Results ①The expression rates of MMP-9 protein and CD133 protein were 40.4%(44/109) and 46.8% (51/109) in 109 cases of breast cancer respectively.The expression of MMP-9 was related with tumor size and TNM stage (P〈0.05).The expression of CD133 was related with tumor grade (P〈0.05).②The expression rates of MMP-9 in CD133+ tumor stem cells(TSC) were related with tumor grades of breast cancer (P〈0.05).Conclusion There is close relationship between CD133+ TSC and MMP-9 in poor differentiation of breast cancer,meanwhile there is a positive correlation between CD133+ TSC and MMP-9 protein in TNM stages.MMP-9 protein in CD133+ TSC can be used as an important factor in judging the prognosis of breast cancer.
