5-溴脱氧尿嘧啶核苷标记骨髓间充质干细胞的技术探讨

摘    要:①目的 探讨5-溴脱氧尿嘧啶核苷(BrdU)标记骨髓间充质干细胞的可行性。②方法 利用密度为1.073g/mL的Percoll分离骨髓的单核细胞,以含10%胎牛血清的低糖DMEM培养基培养与扩增MSCs细胞,纯度可达95%左右。用BrdU标记骨髓间充质干细胞24、48、72和96小时后的检测标记率。③结果 随着标记时间的延长,标记率逐渐增高,标记72小时后标记率在85%以上。④结论 用BrdU标记骨髓间充质干细胞的最佳时间是72小时,最佳浓度为10μg/mL。

Abstract:Objective This study investigated the feasibility of labelling MSCs with BrdU. Methods Bone marrow was aspirated from the rabbit tibia. Mesenchymal stem cells in each bone marrow samples were obtained by gradient centrifugation with Percoll at a density of 1.073g/ml and then cultured and expanded with lower glucose Dulbecco's modified Eagle's medium (L-DMEM) containing 10% fetal bovine serum (FBS). The rate of label was respectively recorded after mesenchymal stem cells were labled with BrdU for 24h ,48h, 72h, and 96h. Results The rate of label increased gradually with time , more than 85% mesenchymal stem cells were labelled after 72h. Conclusion The most appropriate time for labeling mesenchymal stem cells is 72h, the most appropriate concentration is 10ug/ml
