白细胞介素10基因-627位点多态性与早发冠状动脉粥样硬化性心脏病的关系

摘    要:目的探讨白细胞介素10(interleukin 10,IL10)基因-627位点多态性与早发性冠状动脉粥样硬化性心脏病(coronary heart disease,CHD)和血清IL10水平的关系。方法应用聚合酶链反应-限制性片段长度多态性办法,检测CHD患者163例和正常对照者112名IL10基因-627位点多态性,采用ELISA法检测血清IL10。结果IL10基因-627位点基因型和等位基因频率在CHD组和对照组之间差异无统计学意义,X^2值分别为1.9324,1.5703,P〉0.05。按性别分层分析,男性组X^2值分别为1.2708,0.8595,P〉0.05;女。女性组X^2值分别为0.8254,0.7127,P〉0.05。血清IL10在AA型、AC型和CC型之间差异有统计学意义,P〈0.05,但在CHD组和对照组之间差异无统计学意义,P〉0.05。结论IL10基因-627位点多态性与中国人汉族人冠病的易感性无显著关联,但可能影响IL10基因的转录活性。

Abstract:Objective To investigate the relationship between interleukin 10 (IL10) gene-627 polymorphisms and serum IL10 level and early-onset coronary heart disease(CHD). Methods The genotype and allele frequency of IL10 gene-627 site was assayed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). DNA samples were obtained from 163 patients with CHD and 112 controls. Serum IL10 level was detected by ELISA. Results No significant difference was found in the distribution of IL10 genotype and allele frequency between the healthy controls and the patients with CHD; Chi-square values were 1.9324 and 1.5703 respectively,P>0.05. Stratification analyses based on different sex still found no significant difference in the distribution of IL10 genotype and allele frequency between the healthy controls and the CHD patients; the Chi-square values in male groups were 1.2708 versus 0.8595, and in female groups were 0.8254 versus 0.7127, P>0.05. Serum IL10 level showed significant differences among AA genotype, AC genotype and CC genotype, but no significant difference was noted between healthy controls and CHD patients. Conclusion These results suggest that IL10 gene-627 polymorphisms are not associated with an increased risk of CHD, but it might assume a role in IL10 gene expression.
