纳洛酮对血管性痴呆大鼠血浆内皮素的影响

目的:研究纳洛酮对血管性痴呆大鼠血浆内皮素( ET)的影响。方法:采用双侧颈总动脉永久结扎方法制备血管性痴呆大鼠。大鼠随机分为三组:假手术对照组,血管性痴呆组,血管性痴呆纳洛酮治疗组(每天0 .8mg/kg纳洛酮腹腔注射,连续7d)。采用非平衡放射免疫法测定大鼠血浆内皮素水平。结果:痴呆组大鼠血浆ET水平( 1 34.77±2 0 .76pg/ml)比假手术组( 74.83±1 4.74pg/ml)显著升高( P<0 .0 1 ) ,纳洛酮组大鼠ET水平( 1 0 2 .61±2 8.5 3pg/ml)虽然也明显高于假手术组( P<0 .0 5 ) ,但比痴呆组大鼠显著降低( P<0 .0 5 )。结论:纳洛酮能够通过降低血管性痴呆大鼠升高的血浆内皮素水平,减轻脑血管过度收缩

Effect of naloxone on plasma endothelin level in rats with vascular dementia

Objective: To study the effect of naloxone on plasma endothelin (ET) level in rats with vascular dementia. Methods : Vascular dementia was established by permanent occulasion of the common carotid arteries. Rates were divided into three groups viz. sham-operated controls, naloxone treated VD rats (naloxone 0.8 mg/kg, ip daily for 7 days), and nontreated VD rats. ET determination was performed with radioimmunoassay. Results:The level of endothelin in the nontreated VD rats (134.77±20.76pg/ml) and the naloxone treated rats (102.61±28.53pg/ml) was significantly higher than that of the sham-operated controls (74.83±14.74 pg/ml, P <0.05). ET level in the naloxone treated VD rats was markly decreased compared to the nontreated VD rats ( P <0.05). Conclusion: Naloxone could decrease the elevated plasma level of endothelin in rats with VD.
