音乐电针对脑出血模型大鼠脑组织病理的影响

摘    要:目的观察音乐电针对脑出血模型大鼠脑组织病理的影响,并探讨其机制。方法将30只清洁级SD成年大鼠随机分为模型组、治疗组和正常组,每组各10只。模型组及治疗组予Ⅱ型胶原酶加肝素钠尾状核注射诱导脑出血大鼠模型,正常组正常喂养。造模后第1 d,治疗组予音乐电针治疗,每日1次,模型组和正常组均正常喂养。14 d后采用Longa法和Berderson法对模型组及治疗组大鼠治疗前后神经功能进行评分。然后处死各组大鼠,取出脑组织,常规脱水、石蜡包埋、组织切片,进行苏木精-伊红(HE)染色及尼氏染色,分别于生物显微镜低倍镜(×100)和高倍镜(×400)下观察,并对尼氏体积分光密度(IOD)定量分析。结果治疗组治疗后Longa、Berderson评分与本组治疗前比较均明显下降(P〈0.05),且均低于模型组治疗后(P〈0.05)。治疗组HE染色及尼氏染色下脑组织病理改变均较模型组轻。模型组尼氏体IOD值低于正常组(P〈0.05)。治疗组尼氏体IOD值高于模型组(P〈0.05)。结论音乐电针疗法对脑出血模型大鼠脑组织具有明显的保护作用和抗损伤修复能力,其作用机制为提高小胶质细胞的抗损伤的积极作用,减轻对正常脑组织的损伤,增进损伤组织的修复,加速坏死组织的吸收;保护和改善神经元尼氏体合成蛋白质的功能,促进神经元修复。

Abstract:Objective To observe the effect of music electro-acupuncture on pathology of brain tissue of rat with model of encephalorrhagia.Methods 30 SD rats were randomly divided into three groups,10 rats in each group.Rat model with of encephalorrhagia were established by caudate nucleus injections of Collagenase Ⅱand heparin in model group and treatment group.Modeling after 1 d music electro-acupuncture was applied in treatment group,once a day,the model group and normal group were normal feeding.Neurological function was evaluated by Longa and Berderson Methods in model group and treatment group before and 14 days after treatment.Brain tissue of model rat were observed by biological microscope after conventional dehydration,embedded in paraffin,tissue sections,HE staining and Nissl staining.Nissl volume IOD was quantitatively analyzed.Results Longa,Berderson score after treatment were significantly decreased in comparison with before treatment in treatment group(P0.05),and were lower than the model group after treatment(P0.05).Brain tissue pathological changes under HE staining and Nissl staining in treatment group were less than that in the model group.IOD values of Nissl volume in model group was lower than that in the normal group(P0.05).IOD values of Nissl volume in treatment model group was lower than that in model group(P0.05).Conclusion Music electro-acupuncture has obvious prevention,anti-injury and repair capacity of brain tissue of rat with encephalorrhagia.Its mechanism may be to increase the positive role of anti-injury of microglia,to reduce damage to normal brain tissue and enhance repair of damaged tissue to accelerate the absorption of necrotic tissue;protect and improve the Nissl bodies of neurons function of protein synthesis to promote neuronal repair.
