黏膜给药新方法研究

目的：探索黏膜给药新方法——电致孔透黏膜给药。方法：用电致孔给药仪透直肠黏膜给药．与被动扩散给药相比较，通过双室扩散池法、HPLC法检测药物中指标成分栀子苷透过量的变化，摸索适宜的电学参数条件。结果：电致孔条件下栀子苷的累积透过量明显高于被动扩散栀子苷的累积透过量，且栀子苷的累积透过量在电学参数波形为方波、脉冲次数为360个时，透过量增大显著。结论：电致孔透黏膜给药方法可行，优化的电致孔条件应用于中药有效成分透直肠黏膜给药有良好的前景。

Study on New Method of Delivering Drug on Rectum Mucous Membrane

Objective： To explore a new method of dehvering drug on rectum mucous membrane by eletroporation. Method： The delivering drug on rectum mucous membrane by eletroporation was studied and compared with passive diffusion, in Franz diffusion cell for isolated pig rectum mucous membrane of geniposide by different electrical parameters of electroporation equipment, respectively. The content of geniposide permeated on rectum mucous membrane of pig was determined by HPLC for groping optimum condition of electrical parameters. Result： The accumulative permeation of geniposide was correlated with electrical parameters, different electrical parameters showed the significant effect on geniposide＇s rectum mucous membrane delivery. The optimum electrical parameters were shap of wave ： square wave, number of pulses： 360. The permeation increased significantly compared with the passive diffusion. Conelusion： The new method of delivering drug on rectum mucous membrane by eletroporation is practicable. Optimum parameters of eletroporation have an favorable future for active constituents in Chinese materia medica to be rectum mucous membrane deliveried.
