黄芪和当归注射液对兔肾缺血再灌注损伤时ATP酶的影响

摘    要:目的:观察黄芪、当归注射液在肾缺血再灌注(IR)损伤中的作用及机制。方法:将33只健康成年日本大耳白兔,随机分为手术对照(control)组、单纯肾缺血再灌(IR)组、黄芪注射液处理IR组(黄芪+IR)和当归注射液处理IR(当归+IR)组。肾缺血1h再灌注48h,取肾作电镜检查;测血清肌酐(Cr)、肾组织中ATP酶活性。结果:IR组肾组织变性改变显,黄芪、当归组病变较IR组明显减轻。与对照组相比,IR组血清Cr的含量升高(P<0.05),肾中ATP酶活性降低(P<0.05);黄芪+IR组,当归+IR组较IR组血清Cr含量降低(P<0.05),肾中ATP酶活性升高(P<0.05),同对照组差异无显性(P>0.05)。黄芪+IR组与当归+IR组血清Cr含量及肾中ATP酶活性差异无显性(P>0.05)。结论:黄芪和当归可能通过保护ATP酶而减轻IR肾损伤。

Abstract:Object: To investigate the effects of Astragalus membranaceus (AM) and Angelica sinensison (AS) injection on rabbit kindey with ischemia reperfusion and explore its mechanism. Methods: Thirty-three adult Japanese white rabbits were randomly divided into the operational control group (Control group), IR group, AM-treated group (AM+IR group) and RAS-treated group (AS+IR group). After renal ischemia 1 hours and reperfusion for 48 hours, the renal tissue were detected under electron microscope. The content of serum creatinine (Scr) and activity ofo ATPase in the kindey were measured. Results: The degenerative changes were remarkable in renal tissue in IR group, but the changes in AM+IR group and AS+IR group were slightly. Compared with Control group, the content of Scr in IR group increased, and the activity of ATPase in renal tissue decreased (P<0.05); Compared with AM+IR group and AS+IR group, the content of IR group decreased, the activity of ATPase in renal tissue increased (P<0.05). There were no remarkable differences of the content of Scr and the activity of ATPase in renal tissue between AM+IR group, AS+IR group and Control group (P>0.05). Conclusion: AM and AS can alleviate the renal ischemia reperfusion injury by protect the activity of ATPase.
