bFGF生物海绵对脊髓损伤神经保护作用的研究Ⅰ

摘    要:目的:探索碱性成纤维细胞生长因子(bFGF)对大鼠脊髓损伤的神经保护作用。方法:用大鼠脊髓半横断损伤模型,对局部脊髓组织和运动皮持进行神经纤维和神经细胞的定量分析。结果:术后1、2、3周bFGF组脊髓切口处神经纤维均较对照组组密,术后1、2周,神经纤维密度测定结果显示bFGF组较对照组明显增加(P<0.05)。对照组运动皮质内锥体层神经细胞肿胀,尼氏体淡染,出现明显的噬神经细胞现象,bFGF组 偶见此现象,对照组胶质细胞记数明显多于bFGF组(P<0.05)。结论:bFGF生物海绵对大鼠受损脊髓局部及投射纤维起源脑区-运动皮质具有明显的神经保护和神经纤维再生作用。

Abstract:OBJECTIVE:To investigate the neuroprotective effect of bFGF following spinal cord injury in rats METHODS:The model of spinal injury was made by hemitransection at T 10 Collagen sponges with bFGF or vehicle were applied to lesion sites At the end of 1, 2, 3 weeks after operation animals were histologically assessed for both damaged spinal cords and motor cortexes RESULTS:Regenerative nerve fibers in the lesion sites were thicker and more condensive in bFGF treated rats than in controls at the end of 1, 2, 3 weeks after operation Significantly swelling pyramidal cells, reduced Nissl bodies and pronounced neurophage phenomenon appeared in motor cortexes in controls , and the amount of glial cells was much more in controls than in bFGF treated rats ( P <0 05) Only rare neurophage phenomenon appeared in bFGF treaed rats CONCLUSION:bFGF had significantly neuroprotective and neural regenerative effects on damaged cords and motor cortexes which projected nerve fibers into spinal cords in rats
