| 摘 要： | 目的研究顺铂对人宫颈癌HeLa细胞生长的作用以及对FHIT，Bax，Ki67表达的影响，初步探讨其作用机制。方法采用MTT法测定1μg／ml，5μg／ml，10μg／ml，15μg／ml的顺铂对Helm细胞增殖的影响；流式细胞仪检测1μg／ml，5μg／ml，10μg／ml顺铂作用24h后的细胞凋亡情况；免疫细胞化学法测定用药前后FHIT，Bax，Ki67在宫颈癌HeLa细胞上的表达。结果MTT比色法表明顺铂抑制体外培养的HeLa细胞生长并且呈时效和量效关系（P〈0．05）；流式细胞术测细胞凋亡率显示不同浓度顺铂作用于HeLa细胞24h后，各组细胞亚G1峰与对照组相比差异有统计学意义（P〈0．05）；免疫组化法结果显示顺铂可以使FHIT和Bax的表达上调，而Ki67的表达则下调（P〈0．05）。结论顺铂对宫颈癌HeLa细胞体外生长具有明显抑制作用，能通过促进FHIT和Bax的表达，抑制Ki67的表达来诱导肿瘤细胞凋亡。
