大鼠自发性高血压致病相关基因的初步筛选

摘    要:目的:探讨自发性高血压大鼠(SHR)和正常血压大鼠(WKY)肠系膜动脉和肾脏组织中基因表达的差异.方法:提取成年SHR和WKY的二级肠系膜动脉和肾脏总RNA,利用高通量RNA阵列技术(RNA array)检测SHR和WKY二级肠系膜动脉和肾脏组织基因表达水平不同的基因,再用RT-PCR证实部分基因表达的改变.结果:芯片筛选出38条基因,其中上调基因19条,下调基因19条.经RT-PCR验证胰岛素样生长因子结合蛋白5和原肌凝蛋白6基因上调,锌指蛋白10和硬脂酰辅酶去饱和酶1基因下调.结论:筛选出的差异基因可能是高血压的相关基因,深入研究筛选出的相关基因及其功能,将为进一步探讨高血压病相关的致病基因打下良好的基础.

Abstract:Objective:To evaluate the difference of gene expression between the second-order mesenteric arteries and kidneys of spontaeously hypetensive rat(SHR) and normal rat(Wistar-Kyoto rats,WKY).Methods: The total RNA samples were obtained from second-order mesenteric arteries and kidneys of SHR and WKY.Microarray was used to determine the level of mRNA expression of two groups.The results were confirmed by RT-PCR.Result: Thirty-eight positive genes were screened from microarray,including 19 up-regulated genes and 19 down-regulated genes.Up-regulated genes(insulin-like growth factor binding protein 5,tropomyosin isoform 6) and down-regulated(stearoyl-Coenzyme A desaturase 1,ring finger protein 10) were identified by RT-PCR.Conclusion: These positive genes are the genes related to hypertension.These results are helpful for investigating the genes related to disease in hypertension patient.
