7-二氟亚甲基-5,4,-二甲烷氧基异黄酮对动脉粥样硬化兔血管内皮功能的影响

摘    要:目的:观察7-二氟亚甲基-5,4'-二甲烷氧基异黄酮(dFMG)对动脉粥样硬化兔血管内皮功能的影响。方法:40只雄性新西兰兔随机抽取32只制作动脉粥样硬化模型,剩余8只作为正常对照组,造模60天后用随机区组法将32只高脂模型兔分为模型对照组、dFMG组、洛伐他汀组、金雀异黄素组,分别给予安慰剂、dFMG、洛伐他汀、金雀异黄素每天5 mg/kg治疗30天。造模60天及治疗30天后分别于耳缘静脉采血测定血浆一氧化氮(NO),内皮素-1(ET-1)水平;同时于治疗30天后处死兔子取离体血管环进行乙酰胆碱(Ach)诱导的血管内皮依赖性舒张功能检测。结果:治疗30天后dFMG组与模型对照组比较,血浆NO水平显著升高(P<0.01),ET-1水平显著降低(P<0.01),Ach诱导的血管内皮依赖性舒张功能显著改善(P<0.01);dFMG组与洛伐他汀组比较无统计学意义(P>0.05)。结论:dFMGEN能影响NO和ET-1的释放,改善Ach诱导的血管内皮依赖性舒张功能。

Abstract:Objective To investigate the effects of 7-difluoromethyl-5,4'-dimethoxygenistein(dFMG) on endothelial function in atherosclerosis rabbit.Methods Thirty two New Zealand rabbits were randomly selected from forty male rabbits to make the atherosclerosis model and the others were fed normal rabbit chow as normal control.After a sixty-day feeding of high fat diet,the rabbits were divided into four groups by using randomized block design: model control group(vehicle solvent 5 mg/kg per day),dFMG group(dFMG 5 mg/kg per day),lovastatin group(lovastatin 5 mg/kg per day) and genistein group(genistein 5 mg/kg per day).They were treated for thirty days.The level of nitric oxide(NO),endothelin-1(ET-1)were detected and compared after collecting blood through the lateral ear vein after feeding for 60 days and treatment for 30 days.After the rabbits were sacrificed,the thoracic aortas were isolated in order to measure their endothelium-dependent relaxations evoked by acetylcholine.Results Comparing with model control,dFMG could significantly increase the serum NO concentration(P<0.01),reduce the serum ET-1 concentration(P<0.01),and significantly improve the endotheliumdependent relaxations evoked by acetylcholine(P<0.01).There was no significant difference in these measurements between dFMG group and lovastatin group.Conclusion dFMG could affect the release of NO and ET-1 and improve the endothelium-dependent relaxations evoked by acetylcholine.
