快眼动睡眠剥夺对抑郁模型大鼠行为学及血清COR和ACTH的影响

摘    要:目的通过观察抑郁模型大鼠剥夺快速动眼相睡眠后的行为学及血清皮质醇(COR)和促肾上腺皮质激素(ACTH)含量的变化,探讨睡眠剥夺的抗抑郁机制。方法30只成年雌雄各半的SD大鼠随机分为健康对照组、抑郁模型组和睡眠剥夺组,每组10只。后2组采用孤养和长期不可预见的中等强度应激制作大鼠抑郁模型,睡眠剥夺组又采用小平台水环境法进行72h快眼动睡眠剥夺,比较各组大鼠自主活动及血清COR和ACTH含量的变化。结果抑郁模型组大鼠血清COR和ACTH含量均较健康对照组有显著升高(P<0.01),而睡眠剥夺组大鼠较抑郁模型组血清中COR和ACTH含量均显著下降(P<0.01),而与健康对照组无显著差异(P>0.05)。结论快眼动睡眠剥夺可逆转抑郁模型大鼠的抑郁样行为,其抗抑郁作用与其对下丘脑—垂体—肾上腺系统(HPA轴)的调整相关。

Abstract:Objective To investigate the effects of rapid eye movement sleep deprivation (REM SD) on depressive behaviors and COR/ACTH in rats treated with chronic mild unpredieted stresses, to explore the mechanisms of anti-depressive with sleep deprivation. Method Thirty adult SD rats, 15 females and 15 males, were randomly divided into normal con- trol group, depressive model group and REM SD group. The depression model was built with separating feeding, lonely, long- term unpredictable and middle stimulation stress methods. In REM SD group, the rats were deprived sleep lasting 72 hours with flowerpot technique. Then tile changes of behaviors and the concentration of COR/ACTH were observed and compared. Results Compared with the normal control group, the autonomic activity and the concentration of COR/ACTH were significantly increased in model group( P 〈 0.01 ). While the concentration of COR/ACTH were significantly decreased in REM SD group than model group ( P 〈 0.01 ), and there was nb significant change compared with the normal control group ( P 〉 0.05). Conclusion It suggested that the REM SD could reverse the depressive behaviors of rats, and its anti-depres- sive mechanisms was related to the function of regulating the hypothalamus-pituitary-adrenal axis.
