摘    要:目的:考察复方血脂宁中4种药材提取液对二苯乙烯苷稳定性的影响,探究该复方的配伍规律。方法:采用UPLC考察不同条件下二苯乙烯苷单体及其在复方各药材提取液中含量随时间的变化规律,流动相甲醇(A)-0.1%甲酸水溶液(B)梯度洗脱(0~2 min,10%~25%A;2~9 min,25%~35%A;9~12 min,35%~40%A;12~40 min,40%~90%A),流速0.2 m L·min-1,柱温45℃,检测波长280 nm。利用化学反应动力学方法计算二苯乙烯苷在不同环境下的降解反应动力学参数,表征其半衰期(t1/2)和降解活化能。结果:二苯乙烯苷单体及其在不同药材提取液中的降解均遵循一级反应动力学规律,在p H 8.3条件下,二苯乙烯苷单体的降解速率常数(K)0.037 5 h-1,t1/2=18.48 h,该成分在山楂提取液中的K=0.012 3 h-1,t1/2=56.35 h,其他3种药材提取液也在不同条件下表现出对二苯乙烯苷的促稳作用。结论:复方血脂宁中药材配伍可不同程度地提高二苯乙烯苷的稳定性,其中以山楂效果最为显著,从稳定性角度探究了中药复方配伍的合理性。

Abstract:Objective:To study on the stability of stilbene glucoside in various extracts of medicinal materials from compound Xuezhining,and to explore compatibility rule of this compound. Method:The content of stilbene glucoside changed with time under different conditions was investigated by UPLC.Based on the chemical reaction kinetics,the parameters of degradation kinetics were calculated under different environments.The activation energy and half-time(t1/2) were investigated. Result:The degradations of stilbene glucoside in different conditions were followed the first-order reaction kinetics.The degradation rate constant (K) of stilbene glucoside in aqueous solution and Crataegi Fructus extract were 0.037 5,0.012 3 h-1,and t1/2 were 18.48,56.35 h under pH 8.3,respectively.The other extracts also showed the stabilizing effect on stilbene glucoside under different conditions. Conclusion:The compatibility of Chinese medicines in compound Xuezhining can improve the stability of stilbene glucoside in different degrees.Among them,effect of Crataegi Fructus is the most significant.
