摘    要: 目的建立LC-MS/MS测定人血浆中盐酸舍曲林浓度的方法。方法以盐酸帕罗西汀为内标,采用乙腈-0.1%甲酸溶液(50∶50)为流动相,以Discovery C18色谱柱为分析柱,通过电喷雾电离源(ESI),以选择离子反应监测(SRM)方式进行检测。用于定量分析的离子反应分别为m/z306.0→274.9(盐酸舍曲林)和m/z 330.1→191.9(盐酸帕罗西汀)。结果盐酸舍曲林线性范围0.332~66.400μg·L-1,定量下限为(0.334±0.002)μg·L-1(n=5)。日内、日间精密度(RSD)小于9%,平均回收率大于95%。结论本法专属性强,样品处理方便,灵敏度高,适用于盐酸舍曲林临床药动学研究。

Abstract:OBJECTIVE To establish a LC-MS/MS method for the determination of sertraline hydrochloride in human plasma.METHODS Paroxetine hydrochloride was used as the internal standard.Sertraline hydrochloride was chromatographed by using a Discovery C18 column.The mobile phase consisted of 0.1% formic acide-acetonitrile(50∶50).Electrospray ionization(ESI) source was applied and operated in the positive ion mode.Selected reaction monitoring(SRM) mode with the transitions of m/z 306.0→274.9 and m/z 330.1→191.9 was used to quantify sertraline hydrochloride and the internal standard,respectively.RESULTS The linear calibration curve was obtained over the concentration range of 0.332~66.400 μg·L-1.The detection limit was(0.334±0.002) μg·L-1(n=5).The inter-and intra-day precisions(RSD) were less than 9%.The average recoveries were above 95%.CONCLUSION The method is proved to be robust,covenient,sensitive and suitable for the clinical investigation of sertraline hydrochloride pharmacokinetics.
