何首乌、女贞子等中药煎剂对阿霉素诱导的猪毛囊细胞凋亡的影响

目的　探讨何首乌、女贞子等中药混合煎剂对阿霉素诱导的猪毛囊细胞凋亡的影响。方法　将体外培养的猪毛囊分别经阿霉素、阿霉素 +中药处理 ,显微镜下观察各组毛囊、毛发生长和毛球部形态变化 ,并用末端脱氧核苷酸转移酶介导dUTP缺口末端标记法 (TUNEL)检测各组毛囊中的凋亡细胞。结果　培养第 4天 ,阿霉素组毛囊、毛发生长长度明显低于阿霉素 +中药组 (P <0 .0 1) ;阿霉素组毛囊TUNEL阳性细胞数明显高于阿霉素 +中药组 (P <0 .0 1) ;毛球形态观察 ,2组均出现退行性变化 ,阿霉素组较阿霉素 +中药组明显。结论　何首乌、女贞子等中药煎剂能在一定程度上抑制阿霉素诱导的猪毛囊细胞凋亡 ,减轻阿霉素对毛囊的损伤。

Effect of Chinese herbal medicine on adriamycin-induced apoptosis of pig hair follicle cells in vitro

Objective To investigate the effect of water soluble extracts of certain Chinese herbs on adriamycin-induced apoptosis of pig hair follicle cells.?Methods Pig hair follicles were cultured in vitro to study the adriamycin-induced apoptosis of hair follicular cells and the effect of the herbal medicine on the induced apoptosis. Hair growth and the morphology of hair follicle bulb were examined under microscope; the apoptosis of pig hair follicle cells was assessed by a TUNEL technique.?Results On day 4 of culture, the hair growth was obviously suppressed by the added adriamycin and the apoptosis in hair follicle there was more marked than that in the adriamycin-plus-herbs group (P<0.01). The apoptotic cell count and catagen/tologen follicle count were less in the group with herbs than in that without.?Conclusion Water extracts of some traditional Chinese herbs could partially inhibit the adriamycin-induced apoptosis of pig hair follicle cells and alleviate the damage of hair follicles.
