缺氧缺血性脑病新生猪血浆及脑匀浆中一氧化氮水平与脑损伤的关系

摘    要:目的:研究不同程度缺氧缺血性脑病(HIE)新生猪模型中血浆及脑匀浆一氧化氮(NO)水平与不同程度脑损伤组织病理改变的关系,探讨NO在HIE发病机制中的作用.方法:实验新生猪29只,分为对照组与不同缺氧时间HIE组,用硝酸还原酶法测定血浆及脑匀浆NO水平,观察其相应脑组织病理改变.结果:不同缺氧时间HIE组新生猪在缺氧处理后血浆NO均较缺氧前显著升高(P<0.01);随着缺氧时间的延长,各HIE组血浆NO升幅亦越大(P<0.05).而脑匀浆NO均较对照组升高,其中对照组与缺氧30、60min两组间差异有显著性(P<0.05);而对照组与缺氧90min组间以及缺氧30、60、90min各组间差异不明显(P>0.05).结论:HIE新生猪模型中,在缺氧的最初1 h内,随着缺氧时间的延长,脑组织损伤越严重,且血浆及脑匀浆NO均显著升高.测定血浆NO可间接反映脑损伤程度.

Abstract:Objective To study the relationship between nitric oxide (NO) in plasma and cerebral neat jelly and the pathological changes of the cerebral tissues in the newborn piglet with different severity of hypoxic ischemic encephalopathy(HIE) and to explore the role of NO in the development of HIE. Methods Twenty-nine male newborn piglets were assigned to the different severity of HIE groups[hypoxia 30 min(group B),60 min(group C),90 min(group D)] and the control group(group A). Their plasma and cerebral neat jelly NO levels were detected by nitrate reductase method and the pathological changes of the involved cerebral tissues were examined. Results The plasma NO levels in the piglets with different severity of HIE were significantly elevated after oxygen deprivation (P< 0.01) and the elevation of NO level in each HIE group was increased with prolonged oxygen deprivation (P< 0.05). The cerebral neat jelly NO level in each HIE group was higher than that in group A. There was a significant difference between group A and B or C (P< 0.05) but little difference between group A and D or among group B,C,and D (P >0.05). Conclusions In the newborn piglet model of HIE,the elevation of both plasma and cerebral neat jelly NO levels significant and the cerebral injury is deteriorated with prolonged oxygen deprivation within the first one hour of the onset of deprivation. Plasma NO levels can indirectly reflect the extent of the cerebral injury.
