胶原蛋白海绵生产中的病毒灭活工艺效果观察

摘    要:目的研究胶原蛋白海绵生产中病毒灭活工艺及其灭活病毒的效果。方法采用细胞培养法,对酸性酶解溶液和60Co-γ射线辐照法灭活病毒的效果进行了检测。结果酸性酶解溶液处理2 h,对接种在胶原蛋白海绵中的水疱性口炎病毒、伪狂犬病毒、脊髓灰质炎病毒和猪细小病毒灭活对数值分别为5.54、5.71、6.30和5.13。用照射剂量为25 kGy的60Co-γ射线对胶原蛋白冻干品中的水疱性口炎病毒、伪狂犬病毒、脊髓灰质炎病毒和猪细小病毒灭活对数值分别为6.00、5.75、6.00和5.00。灭活后的样品经细胞感染盲传3代均未见细胞病变。结论胶原蛋白海绵生产中采用酸性酶解溶液处理72 h或25kGy60Co-γ射线辐照灭菌工艺均可有效灭活所试验的4种指示病毒。

Abstract:Objective To study the virus inactivation effect and virus inactivation technique in the production of collagen protein sponge.Methodd Cell culture method was used to observe the virus inactivation effect of acid solution and 60Co-γ radiation 25 kGy.Resultd The average killing logarithm values of vesicular stomatitis virus,pseudorabies virus,poliomyelitis virus and swine parvovirus polluted in collagen protein sponge exposed to acid solution for 2 hours were more than 5.54,5.71,6.30 and 5.13 respectively.The average killing logarithm values of vesicular stomatitis virus,pseudorabies virus,poliomyelitis virus and swine parvovirus in the samples of freeze-drying collagen exposed to 60Co-γ radiation 25 kGy were more than 6.00,5.75,6.00 and 5.00 respectively.No CPE was found in the cell line which was inoculated with treated samples after blind passage for three generations.Conclusion Acid solution treatment for 72 hours and 60Co-γ radiation 25 kGy which are used in the production of collagen protein sponge have good effects on virus inactivation.
