摘    要:本文对提高麦迪霉素有效组分A_1作了研究。根据麦迪霉素有效组分A_1与其主要杂质柱晶白霉素A_6结构上的区别,在发酵培养基中流加了能转化为丙酰CoA的前体物质,得到了较为满意的结果。实验证明,流加蛋氨酸,适当调整培养基中的葡萄糖、玉米粉含量,在不影响效价的情况下,可使有效组分A_1的含量提高8.7%。使用正交设计法,得到最佳条件的组合,使有效分组A_1的含量提高9.7%。流加异亮氨酸、缬氨酸可使有效组分A_1的含量提高13%和10.2%。

Abstract:Based on the different structure of the main component of Midecamycin, Midecamycin A_1 and the less therapeutic efficay one, Leucomycin A_6,a fermentation process development to increase Midecamycin A_1 by continuous adding of precursor which could transform propion-yl coenzyme A was studied. Results showed that by adding of methionine and modifying the composition of the fermentation medium such as glucose and corn starch meal, which was not relatively lowered the titer of the antibiotic produced, the main component, Midecamycin A_1, could be increased by 8.7%. By the aid of orthogonal design method, the optimum composition of the medium was found, and the content of Midecamycin A_1 was further increased to 9.7%. When isoleucine or valine was added as precursor, the content of Midecamycin A_1 could reach 13% and 10.2% respectively.
