摘    要:应用明胶底物膜方法测定了4′-羰基乙氧苯基4-胍基苯甲酸酯盐酸盐(CEGB)对生育男子精子的明胶水解(顶体蛋白酶)活性的抑制作用。实验结果表明当底物膜中 CEGB 的浓度逐渐增加时,精子的晕圈形成率(SHF)逐渐减少,平均晕圈直径(MHD)和平均溶解面积(MLA)都逐渐缩小;当CEGB 的浓度增加到6μmol/L 时则可完全抑制精子的晕圈形成。结果表明 CEGB 能够抑制人精子的明胶水解活性。

Abstract:In vitro study was performed to evaluate the effect of 4′-carboethoxyphenyl 4-guanidino benzoate hy- drochloride(CEGB) on acrosin activity of human sperm.A modified gelatin substrate film assay was used to test the acrosin activity,marked by a halo for- mation around the sperm head.A positive correlation exists between the size of the halo,the lysis area(halo formation) and the degree of acrosin activity.Infiltra- tion of the gelatin substrate film with proteinase in- hibitor CEGB in concertrations of 2,4,6,8,10/μmol/L before adding sperm suspension showed reduction in number of halo formations and in diameter of halo in different degrees.Halo formation was completely sup- pressed when CEGB was present at s concentration of 6μmol/L.The results indicated that a negative correla- tion exists between the concentration of CEGB and the acrosin activity(r=-0.995).
