| 摘 要： | 目的 分析构成关节软骨钙化层有机成分中胶原蛋白类型与含量及无机成分中羟基磷灰石含量.方法 组织库获取20例自愿捐献的人体正常股骨髁新鲜标本,组织切片采用天狼猩红染色和免疫组织化学对胶原蛋白进行定性分析;用氨基酸分析和X-射线衍射法分别对胶原蛋白和羟基磷灰石进行定量分析.结果 天狼猩红染色见钙化软骨层与透明软骨层一样黄绿色淡染、弱双折光性;Ⅱ型胶原蛋白免疫组织化学分析,钙化软骨层呈强绿色荧光,潮线附近的胶原蛋白呈板层平行分布;氨基酸定量分析钙化软骨层Ⅱ型胶原蛋白含量占组织干质量的(20.16%±0.96%);X-射线衍射定量分析钙化软骨层中的羟基磷灰石含量占无机成分的(65.09%±2.31%).结论 在关节骨软骨复合组织中,钙化软骨层是位于透明软骨层与软骨下骨之间的界面连接结构,其细胞外基质主要由Ⅱ型胶原蛋白和羟基磷灰石组成.
