小鼠乳腺癌细胞4T1对不同品系小鼠乳腺癌模型的影响

摘    要:目的:将小鼠乳腺癌细胞4T1接种到两种不同品系的小鼠脂肪垫,观察植瘤小鼠的生物学状态、成瘤大小、转移情况以及生存周期等指标,为建立不同品系小鼠的乳腺癌模型提供参考。方法:将小鼠乳腺癌细胞4T1胰酶消化去除上清,用PBS洗两遍,最后用生理盐水调整细胞数量为1×106个,分别接种到裸鼠和BABL/c小鼠的脂肪垫。比较各组老鼠的成瘤率、成瘤时间以及生存期等实验指标。结果:原位注射1×106个4T1细胞时两组均能成瘤,但是裸鼠的成瘤率高于BABL/c小鼠;裸鼠的成瘤大小明显大于BABL/c小鼠;裸鼠的生存期明显短于BABL/c小鼠;裸鼠出现肺转移结节明显多于BABL/c小鼠。结论:不同免疫能力的小鼠对接种相同数量的细胞数可能会产生不同的实验结果,在选择构建小鼠乳腺癌模型的时候一定要综合考虑所选用品系的小鼠是否会对实验结果带来影响。

Abstract:Objective: To establish different types of breast cancer animals models by observing the biological status of tumor-bearing mice, tumor size, metastasis, survival and other indicators. Methods: The mouse breast cancer cell line 4T1 was trypsinized and the supernatant was removed, washed twice with PBS, and finally 1×106 cells were adjusted with physiological saline and injected into the fat pads of nude mice and BABL/c mice, respectively. The tumor formation rate, tumor formation time and survival time of mice in each group were compared. Results: In situ injection of 1×106 4T1 cells could induce tumor formation in both groups, but the tumor formation rate in nude mice was higher than that in BABL/c mice; the tumor formation size in nude mice was significantly larger than that in BABL/c mice. The survivals of BABL/c mice was significantly shorter than those of BABL/c mice. The number of lung metastatic nodules in BABL/c mice was significantly higher than that of BABL/c mice. Conclusion: Mice with different immune capacity may produce different experimental results when injected with the same number of cells. When building a mouse breast cancer model, it is necessary to consider that different types of mice may bring different experimental results.
