ET、CK-18 M30在大鼠非酒精性脂肪性肝炎组织中的表达及意义

摘    要:目的 探讨血清内毒素(ET)和细胞角蛋白-18 M30(CK-18 M30))在大鼠非酒精性脂肪性肝炎(NASH)中的表达及意义。方法 20只雄性SD大鼠,完全随机配对将6只分配到观测组,14只按体重配成区组并随机分为对照组和实验组。对照组饲以基础饲料,实验组与观测组饲以高脂饲料。每隔4周从观测组随机选取2只大鼠,观察肝组织病理学改变是否达到NASH,若达到,处死对照组和实验组全部大鼠。测定血清中谷丙转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、γ谷氨酰转移酶(GGT)、甘油三酯(TG)、低密度脂蛋白(LDL)、总胆固醇(TC)、CK-18 M30、ET,肝组织进行石蜡包埋切片HE染色。结果 12周末大鼠病理评分、肝指数、血清ALT、TG、LDL、GGT、CK-18(M30)、ET水平均高于对照组(P<0.05),体重低于对照组(P<0.05),AST、GGT水平高于对照组,但差异无统计学意义(P>0.05);spearman相关性分析显示ALT与病理评分无相关性(P>0.05);血清TG、LDL、TC、CK-18 M30及ET与肝脏病理评分呈正相关(P<0.05)。CK-18 M30+ET的AUC面积为1.000。CK-18 M30+ET的敏感度100%、特异度100%。结论 血清ET与CK-18 M30水平联合检测有望成为NASH新的无创检测方式。

Abstract:Objective To investigate the expression and significance of serum endotoxin (ET) and cytokeratin-18 M30 (CK-18 M30) in rat nonalcoholic steatohepatitis (NASH).Methods Twenty male Sprague-Dawley rats were randomly assigned to 6 rats in the observation group,and 14 were randomly divided into control group and experimental group.The control group was fed with basic diet,and the experimental group and the observation group were fed with high fat diet.Two rats were randomly selected from the observation group every 4 weeks to observe whether the pathological changes of liver tissue reached NASH.If they were reached,all the rats in the control group and the experimental group were sacrificed.Determination of serum alanine aminotransferase (ALT),aspartate aminotransferase (AST),gamma glutamyl transferase (GGT),triglyceride (TG),low density lipoprotein (LDL),total cholesterol (TC),CK-18 M30,ET,liver tissue was stained with paraffin-embedded sections for HE staining.Results Twenty male Sprague-Dawley rats were randomly assigned to 6 rats in the observation group,and 14 were randomly divided into control group and experimental group.The control group was fed with basic diet,and the comexperimental group and the observation group were fed with high fat diet.Two rats were randomly selected from the observation group every 4 weeks to observe whether the pathological changes of liver tissue reached NASH.If they were reached,all the rats in the control group and the experimental group were sacrificed.Determination of serum alanine aminotransferase (ALT),aspartate aminotransferase (AST),gamma glutamyl transferase (GGT),triglyceride (TG),low density lipoprotein (LDL),total cholesterol (TC) ,CK-18 M30,ET,liver tissue was stained with paraffin-embedded sections for HE staining.Conclusion The combined detection of serum ET and CK-18 M30levels is expected to become a new non-invasive detection method for NASH.
