中药金叶败毒制剂对人巨细胞病毒感染细胞kip1基因转录水平的影响

摘    要:目的 研究中药金叶败毒制剂对人巨细胞病毒(HCMV)感染细胞细胞周期调控抑制基因kip1转录水平的影响,探讨中药金叶败毒制剂体外对HCMV感染的阻断机制.方法 2005年7月至12月于南方医科大学南方医院,采用逆转录聚合酶链法(RT-PCR)检测HCMV感染后24h、48h、72h、96h细胞中kip1 mRNA水平,检测中药金叶败毒制剂干预后kip1 mRNA水平的变化.结果 病毒感染后24h、48h、72h、96h细胞中kip1 mRNA水平呈下降趋势,而中药金叶败毒制剂可部分阻止这种趋势.结论 中药金叶败毒制剂部分上调HCMV感染引起的kip1 mRNA水平下降,可能是中药金叶败毒制剂抗HCMV效应的作用机制之一.

Abstract:Objective The effects of traditional herb Jinyebaidu on the levels of control-inhibiting gene kipl mRNA variation of concentration in the cell cycle of infected HCMV cells; moreover , the block mechanisms of Chinese traditional herb Jinyebaidu against virus infection cell in vitro were investigated. Methods The kipl mRNA levels of the infected HCMV cells after 24h,48h,72h and 96h were measured by RT-PCR, and the change of the kip1 mRNA levels interfered by Chinese traditional herb Jinyebaidu was detected. Results The kipl mRNA levels of infected HCMV cells showed the trend of decreasing from 24h to 96h, but Chinese traditional herb Jinyebaidu could partly arrest this trend. Conclusion Chinese traditional herb Jinyebaidu can partly up-regulate the kipl mRNA levels in HCMV infected cells, therefore it may be one of the antivirus mechanisms.
