甲基叔丁基醚的蓄积毒性和遗传毒性研究

摘    要:选择中国及德国产两种MTBE样品,进行小鼠蓄积毒性、微核、Ames、精母与精原细胞染色体畸变试验。结果表明,MTBE蓄积系数大于5。小鼠经呼吸道或经口染毒后,染毒剂量达1/2LD50或LC50时,与对照组比较,微核率仍无显著性升高,也未见明显剂量反应关系。Ames试验中,1、10、100、1000μg/皿剂量下,加与不加S9,4个受试沙门氏菌株均未见诱变性。但精母细胞性染色体早熟分离及精原细胞多倍体发生率显著升高(P<001)。由此可见,MTBE属轻度蓄积物质。微核及Ames试验均为阴性。但引起小鼠精母细胞性染色体早熟分离及精原细胞多倍体发生率显著增加的机制与意义,值得进一步探讨

Abstract:In order to study the accumulative and genetic toxicities of MTBE.Two MTBE samples(produced in China and German)were studied with accumulation toxicity test,micronucleus test,Ames test,spermatocyte and permatogenous cell chromosome aberration test.Results show that the accumulation coefficient of MTBE through inhalation in mice is lager than 5, no genetic toxicities could be found in micronucleus test or in Ames test.However,the occurring rates of the early maturing sepration of spcrmatocyte allosomes and the polyploid of permatogenous cells are both significant higher than the controls'(P<0 01).Although MTBE belongs to a low accumulative substance.But,a further study to determine whether MTBE has a potential genetic toxicity to reproductive cells is necessary.
