宫颈鳞状细胞癌组织中Bcl-2与血管内皮生长因子的表达及相关性

摘    要:目的探讨凋亡抑制基因Bcl-2与血管内皮生长因子(VEGF)蛋白在宫颈鳞状细胞癌中的表达,以及二者表达与宫颈鳞状细胞癌中生物学行为的相关性。方法分别对45例宫颈鳞状细胞癌中组织、34例CIN组织和10例正常宫颈组织,采用免疫组织化学法检测Bcl-2的表达,免疫蛋白印迹(Western Blotting)法检测VEGF蛋白的表达,并采用DNA原位末端标记(TUNEL)法检测细胞凋亡指数(AI)。结果 Bcl-2和VEGF蛋白在45例宫颈鳞状细胞癌组织中的阳性表达率分别为51.11%和62.22%,均显著高于CIN组织(11.76%和5.88%)(P<0.05);而在正常宫颈组织二者均无表达。同时,Bcl-2在VEGF表达阳性的宫颈鳞状细胞癌组织中呈高度表达,Bcl-2和VEGF的表达密切相关(χ2=5.148,P<0.05)。结论提示Bcl-2可能参与宫颈鳞状细胞癌的发生和发展;Bcl-2可能通过上调VEGF的表达而促进宫颈鳞状细胞癌的转移,同时VEGF也可能通过诱导Bcl-2的表达而抑制肿瘤细胞的凋亡。

Abstract:Objective To investigate the expression of significance of bcl-2 as a member of inhibitor apoptosis gene family and its correlation with vascular endothelial growth factor (VEGF) protein in the tissues of squamous cell carcinoma of the cervical. Methods Immunohistochemical techniques were used to detect the expression of bcl-2 and Western Blotting technique was used to detect the expression of VEGF protein in 45 cases of squamous cell carcinoma of the cervical tissues, 34 cases of CIN tissues and 10 cases of normal cervical tissues, respectively. The apoptosis index (AI) was detected by TUNEL method. Results The positive expressions of bcl-2 and VEGF protein in squamous cell carcinoma of the cervical tissues were respectively 51.11% and 62.22% which were significantly higher than CIN tissues(P0.05). There was no experession in normal cervical tissues. The expression of bcl-2 had no significant correlation with metastasis of squamous cell carcinoma of the cervica and the degree of differentiation. There was a significant correlation between the high expression of bcl-2 and VEGF (χ2=5.148, P0.05). Conclusion The high expression of bcl-2 in squamous cell carcinoma of the cervica suggests that it may play an important role in the mechanism of genesis and development of squamous cell carcinoma of the cervica. The correlation between the expression of bcl-2 and VEGF indicate that bcl-2 may promote the metastasis of squamous cell carcinoma of the cervica through up-regulating the expression of VEGF, meanwhile, VEGF might inhibit the apoptosis of neoplastic cell by inducing the expression of bcl-2.
