人巨细胞病毒负荷量对细胞病变的影响

摘    要:目的探讨感染后人巨细胞病毒负荷量与细胞病变的关系。方法采用不同滴定度的人巨细胞病毒感染传代培养的人胚肺成纤维细胞,建立人巨细胞病毒梯度感染细胞模型,荧光定量PCR法测定感染细胞内及其维持液病毒DNA量,光镜观察致细胞病变作用,电镜检查其超微结构的改变。结果人巨细胞病毒感染后,细胞内与其维持液病毒DNA量成正相关(r=0.83,P<0.01)。当细胞内病毒DNA量>105GE/106HEL时,病毒开始自细胞内释放;当维持液病毒DNA量>103GE/ml时,细胞出现病变。结论人巨细胞病毒感染后,受染细胞排放病毒致细胞外,且细胞内外病毒负荷量成正相关。细胞病变的发生情况与细胞内及其周围环境的病毒负荷量均密切相关。

Abstract:Objective To examine the relationship between HCMV infection and cytopathic effect (CPE) in human embryonic lung cells (HEL). Method HEL cells were infected with various concentrations of HCMV. The levels of viral DNA were quantified by FQ-PCR. CPE was determined by microscopice observation of the infected cell culture. The ultra-structural changes were studied by TEM. Result After HCMV infection, a positive correlation between the quantity of intracellular and supernatant HCMV DNA was observed (r=0.83, P<0.01). When the level of intracellular HCMV DNA was greater than 105 GE/106 HEL, virions were released from the infected cells. CPE was observed when the level of viral DNA in culture supernatant reached to a level of greater than 103 GE/ml. Conclusion A positive correlation between intracellular virus loading and the release of virions into culture supernatant was observed. The development of CPE is correlated with the quantity of viral DNA in the infected HEL.
