摘    要:目的探讨补骨脂素(PSO)加长波紫外线(UVA)光化学疗法(PUVA)诱导人白血病细胞K562凋亡时对Fas表达的影响。方法以K562细胞为研究对象,以细胞凋亡率、电镜下细胞超微结构改变以及细胞Fas基因表达水平、蛋白表达水平为检测指标,观察中药补骨脂中提取的PSO加波长为360 nm的UVA对人白血病细胞诱导凋亡的作用以及部分作用途径,并采用多因素方差分析法进行统计学处理。结果(1)单纯PSO、单纯UVA照射及PUVA均可诱导K562细胞发生凋亡,PUVA的作用显著强于前两者。(2)电镜下观察经PUVA处理后的K562细胞超微结构,可见明显的凋亡形态学改变。(3)单纯PSO、单纯UVA照射及PUVA均可上调K562细胞Fas基因表达水平,PUVA的作用显著强于前两者。(4)单纯PSO、单纯UVA照射及PUVA均可上调K562细胞Fas蛋白表达水平,PUVA的作用显著强于前两者。结论(1)PUVA可诱导白血病细胞K562发生凋亡,作用强于单纯PSO及单纯UVA照射。(2)PUVA诱导K562凋亡的途径之一为上调K562细胞Fas基因表达水平。
