结肠癌组织中存活素、PTEN、p53基因表达检测

目的:检测结肠癌组织中存活素(Survivin)、PTEN和p53基因的表达,并分析其与结肠癌发生发展、临床病理特征之间的关系.方法:应用免疫组织化学SP法检测59例结肠癌、20例结肠腺瘤、19例正常结肠组织中Survivin、PTEN和p53基因的表达,并结合患者的临床病理资料进行分析.结果:结肠癌组织中Survivin、PTEN、突变型p53的阳性表达率分别为64.4%、54.2%、66.1%,Survivin和突变型p53在正常结肠组织和结肠腺瘤组织中不表达, PTEN基因在正常结肠黏膜和结肠腺瘤组织中几乎全部呈阳性表达.3项指标在不同组织中的表达阳性率差异均有统计学意义(P均＜0.05).结肠癌组织中Survivin和突变型p53的阳性表达率均显著高于结肠腺瘤以及正常结肠黏膜组织 (P＜0.05).随结肠癌组织学类型的变化、淋巴结的转移, Duck's分期的增加,突变型p53的阳性表达率逐渐升高(P＜0.05), PTEN的阳性表达率逐渐降低(P＜0.05).Survivin蛋白在结肠癌组织中的表达与其临床病理参数无关.在结肠癌组织中Survivin蛋白与突变型p53蛋白的表达呈正相关((r=0.813 5,P＜0.05), 与PTEN蛋白的表达呈负相关(r=-0.773 5, P＜0.05), PTEN蛋白与突变型p53蛋白的表达呈负相关(r=-0.935 0,P＜0.05).结论:Survivin、PTEN和p53蛋白的异常表达与结肠癌的发生、发展和病理学特征有关.

Detection of survivin, PTEN, and p53 gene in colon carcinoma tissue

Aim:To detect expressions of antiapoptosis gene survivin,anti-oncogene PTEN and p53,and their relationship with tumor progress and clinical pathological characteristics in colon carcinoma tissue.Methods:The expressions of survivin,PTEN and p53 were examined by immunohistochemistry S-P in 59 cases of colon carcinoma,20 cases of enteric adenoma,and 19 cases of normal colon tissue.Results:The positive rates of survivin,PTEN,and mutant p53 were 64.4%,54.2% and 66.1% in colon carcinoma tissue.The survivin gene and mutant p53 gene were not expressed in normal colon tissue or enteric adenoma tissue.PTEN gene was expressed in almost all the normal colon tissue and enteric adenoma tissue.The positive rates of survivin gene and mutant p53 gene in colon carcinoma tissue were higher than those in normal colon tissue and enteric adenoma tissue.The positive rate of mutant p53 increased while that of PTEN decreased with the increase of Duck's stage,changes of histological type,and lymph node metastasis(P<0.05).In colon carcinoma tissue,the expression of survivin was positively correlated with that of mutant p53(r=0.813 5,P<0.05),and negatively correlated with PTEN(r=-0.773 5,P<0.05).The expression of PTEN was negatively correlated with that of mutant p53(r=-0.935 0,P<0.05).Conclusion:Abnormal expressions of survivin,PTEN,and mutant p53 gene are related to tumor malignant progress and pathological characteristics of colon carcinoma.
