二期梅毒患者外周血Th1/Th2及Tc1/Tc2的研究

摘    要:目的探讨CD4+T细胞(Th细胞)和CD8+T细胞(Tc细胞)亚群在梅毒发病机理中的作用及其相互关系。方法采用流式细胞术检测经三色荧光抗体染色的二期梅毒患者外周血CD4+/IFN-γ+(Th1),CD4+1/IL-4+(Th2),CD4+/IFN-γ+/IL-4+(Tho),CD8+/IFN-γ+(Tc1),CD8+/IL-4+(Tc2),CD8+/IFN-γ+/IL-4+(Tc0)细胞含量。结果梅毒组Th1细胞含量、Th1/Th2,Tc1/Tc2比值均明显低于正常对照组(P<0.01),而Tc0和Tc2细胞含量显著高于正常对照组,其他细胞两组间无显著性差异(P>0.05)。结论二期梅毒患者Th1/Th2及Tc1/Tc2比值失衡,可能是机体不能完全清除梅毒螺旋体造成其潜伏体内形成长期感染的重要因素之一。

Abstract:Objective To investigate the pattern of cytokines produced by CD_4~+ and CD_8~+ T-lymphocytes and to understand the role of Th and Tc lymphocyte subsets in the pathogenesis of secondary syphilis and the relationship between them.Methods A three-colour immunoflurescent antibody to stain the peripheral blood T cells in study group and the controls and to detect CD_4~+ /IFNγ~+ (Th1),CD_4~+ 1/IL-4~+ (Th2),CD_4~+ /IFNγ~+/IL-4~+ (Tho),CD_8~+ /IFNγ+ (Tc1),CD_8~+ /IL-4~+ (Tc2),CD_8~+ /IFNγ~+/IL-4~+ (Tc0) cells was adopted by flow cytometry.Results Compared to that of controls,patients with secondary syphilis showed a decreased number of Th1 cells and a decreased rate of Th1/Th2 and Tc1/Tc2,and a increased number of Tc2 and Tc2,but there was no difference in number of Th2,Th0 and Tc1 cells between the two groups (P>0.05).Conclusion It showed that differentiation disorder in peripheral blood Th cells and decreased rate of Th1 cells and increased rate of Tc2 may take an important role in the mechanism of secondary syphilis.
