沙樱桃黄酮对人A-704细胞毒性及增殖的影响

摘    要:目的探讨沙樱桃黄酮对人A-704(肾癌)细胞毒性作用及其对细胞增殖的影响,为沙樱桃的应用和开发提供实验依据。方法体外培养人A-704细胞沙樱桃黄酮,用台盼蓝法检测对细胞毒性作用;用克隆形成法测定对细胞分裂增殖的影响;用X射线能量色散谱仪分析对细胞内微量元素含量的影响。结果①台盼蓝法测定结果显示,给药后培养6h,沙樱桃黄酮(0.1,0.5,5mg.L-1)组和5-氟尿嘧啶(5-FU)组活细胞数比对照组分别减少了33.8%、35.8%、60.8%和40.3%,到18h减少了57.3%、80.1%、99%和59.6%,差异有显著性(P<0.01)。沙樱桃黄酮组间比较,差异有显著性(P<0.01)。②克隆形成法测定结果表明,沙樱桃黄酮(0.1,0.5,5mg.L-1)组和5-FU组克隆数比对照组减少了40.4%、47.6%、83.6%和43.4%,差异有显著性(P<0.01)。③X射线能量色散谱仪分析结果表明,细胞内Ca2+离子水平随给药浓度的增加而升高,K+和P3+离子浓度则降低,与对照组比,差异有显著性(P<0.01)。结论沙樱桃黄酮对人A-704细胞具有明显的毒性作用,其作用与抑制细胞分裂,改变细胞膜结构,使细胞内游离Ca2+增加活化细胞内核酸内切酶,引起细胞DNA损伤有关。

Abstract:Aim To observe the cytotoxic and proliferous effects of Nitraria tangutorum flavone(NTF)on human A-704 strain cells.Methods In vitro cultured human A-704 strain cells,the cytotoxic and proliferous effects of NTF on human A-704 strain cells were determined with methods of trypan-blue and clone formation,and the microelement in cell was detected with X-electron microscope scanning.Results ① The trypan-blue detection showed that the living cells in the NTF group(0.1,0.5 and 5 mg·L-1)and the 5-FU group were decreased by 33.8%~57.3%,35.8%~80.1%,60.8%~99% and 40.3%~59.6% at 6~18 hour after treatment,which significantly differed from those of the control group(P<0.01);there were significant differences between the NTF groups(P<0.01).② The clone formation results showed that the clones in NTF group(0.1,0.5 and 5 mg·L-1)and the 5-FU group were decreased by 40.4%,47.6%,83.6% and 43.4%,compared to the control group(P<0.01).③ X-electron microscope scanning showed that the Ca2+ was increasing and the K+,P3+was decreasing in the cell with the increase of NTF.Conclusion NTF has significant cytotoxic effects on human A-704 strain cells and the mechanism may be related to inhibition of cell division,destruction of structure of cell membrane and change of level of microelement.
