~(99)Tc~m-EGFR-McAb及~(99)Tc~m-CD44-McAb单独及联合用于荷人肺腺癌裸鼠的显像研究

摘    要:目的 评价99Tcm-EGFR-McAb及99Tcm-CD44-McAb单独及联合应用于荷人肺腺癌裸鼠的放射免疫显像效果.方法 建立荷人肺腺癌裸鼠动物模型;采用直接法99Tcm标记抗EGFR和CD44的单克隆抗体,运用柱层析法对标记抗体分离、纯化,并对标记、纯化后的抗体特性进行鉴定;利用99Tcm-EGFR-McAb及99Tcm-CD44-McAb单独及联合进行倚人肺腺癌裸鼠显像及体内分布研究.结果 99Tcm对EGFR-McAb和CD44-McAb的标记率分别为(91.5±3.8)%、(92.3±4.1)%,标记抗体比活度分别为(2.8±0.3)MBq/μl、(2.9±0.5)MBq/μl,放射化学纯度分别为(96.5±2.8)%、(96.2±3.1)%.放射免疫显像结果显示,肿瘤组织对2种标记抗体均有较高的摄取,2种标记抗体联合使用肿瘤部位的放射性浓聚明显高于2种标记抗体单独使用,通过ROI技术测得T/NT值分别为(5.58±0.46)、(2.72±0.22)、(2.30±0.18).标记抗体的体内分布规律与显像结果一致.结论 99Tcm-EGFR-McAb、99Tcm-CD44-McAb用于荷人肺腺癌裸鼠的放射免疫显像能得到较理想的靶/非靶比值,2种标记抗体的联合应用,可在一定程度上提高靶/非靶比值,优于一种标记抗体的单独使用.
