微囊化对转VEGF基因成肌细胞血管内皮生长因子分泌的实验研究

摘    要:目的观察微囊化过程对转VEGF基因成肌细胞分泌VEGF蛋白的影响,从而为微囊化细胞应用于促进缺血皮瓣动物模型皮瓣成活的实验研究做准备。方法将成长旺盛的转VEGF基因成肌细胞分别进行微囊化及非微囊化。14 d内定期对培养液定量提取,检测VEGF蛋白浓度,进行统计学分析。结果检测结果证实,微囊化及非微囊化后无明显差别,两者VEGF浓度均在1~4 d上升,在第5 d接近峰值,7 d后浓度缓慢下降。结论微囊化与否对转VEGF基因成肌细胞分泌VEGF蛋白过程没有显著影响。

Abstract:Objective To observe the influence of microencapsulation process in the courses of myoblasts′ VEGF gene secrete VEGF protein.To prepare for the promotion of animal models of ischemic skin flap with flap survival.Methods The myoblasts were processed by different approaches,microencapsulation and non-microencapsulation.Regular medium quantitative extraction was pdrformed within 14 days.To detect the VEGF protein concentration and the data was analyzed statistically.Results It is no significant difference in the microencapsulated and non-microencapsulated,both the concentration of VEGF increased in 1~4 days near the peak in the first five days,a slow decline in the concentration of 7 days.Conclusion microencapsulated or not has no concert with the VEGF secretion of VEGF transgenic myoblasts.
