大肠癌p53基因突变及p16基因缺失检测及临床意义

摘    要:目的:探讨本地区散发性大肠癌p53、p16蛋白表达、临床病理因素的关系。方法:SSCP-PCR检测p53基因突变、p16基因缺失,免疫组化检测p53,p16蛋白表达,结果p16基因缺失率48%(19/40例),未见突变,p16蛋白缺失率为55%(22/40)。p53基因突变率30%,p53蛋白阳性表达率为68%(27/40例)。组织学分组Ⅱ,Ⅲ级p16基因缺失率明显高于Ⅰ级(P<0.01),临床分期D期p53估变明显高于临床分期B,C期(P<0.05),结论p16基因缺失、53基因突变与大肠癌的发生、发展有关。

Abstract:Objective To study the p16 gene deletion and p53 gene mutation, and their relationship with protein expression and clinicopathological features.Methods p16 gene deletion and p53 gene mutation were detected by SSCP-PCR, and their proteins were measured by immunohistochemical technique in 40 cases of colorectal cancer.Results The p16 gene deletion rate was 48% and no mutation was found. p16 protein expression deletion rate was 55%. p53 gene mutation ration was 30%. p53 protein expression rate was 68%. There were significant difference between grade Ⅱ,Ⅲ and grade Ⅰ in p16 gene deletion, and between dukes D and Dukes B or C in p53 gene mutation. Conclusion p16 gene deletion and p53 gene mutation may be associated with progression of colorectal carcinoma.
