P-gp在人肝硬化及肝细胞肝癌中的表达及意义

目的探讨P糖蛋白(P-gp)在原发性肝细胞癌(HCC)中的表达及其与临床病理特征的关系。方法采用免疫组织化学SABC法分别检测41例HCC、10例肝硬化及10例正常肝组织中P-gp蛋白的表达。结果P-gp在HCC组织中阳性表达率为70.73%(29/41),肝硬化中P-gp阳性表达率为40%(4/10),正常肝组织P-gp阳性表达率为30%(3/10)。3种组织P-gp表达不同(P<0.05);P-gp在正常肝组织与HCC组织中的表达比较差异有统计学意义(P<0.017);在肝硬化与HCC组织中的表达比较差异具有统计学意义(P<0.017);而在肝硬化组织和正常肝组织之间差异无统计学意义(P>0.017)。P-gp的阳性表达与HCC的Edmenson分级明显相关,分化程度越低,P-gp表达越高。结论P-gp蛋白在HCC中高表达,可能参与了HCC的发生、发展及恶性增殖。

Expression of P-glycoprotein in Human Liver Cirrhosis and Hepatocellular Carcinoma

Objective To investigate the expression of P-glycoprotein and to explore the relationship between the P-gp expression and the clinicopathologic features in human liver cirrhosis and hepatocellular carcinoma. Methods The expression of P-gp in 41 cases with HCC, 10 cases with cirrhosis and 10 cases with normal tissue of liver were detected by immunohistochemical technique （streptavidin-biotin complex, SABC）. Results P-gp positive was detected in 29 （70.73%） of 41 HCC ,4 of 10 in cirrhosis ,3 of 10 in normal tissues, positive granute is mainly in cell membrane. There were significant difference in HCC, cirrhosis and normal liver （P 〈0.05 ）. In the area of normal liver,the expression was significant lower than that in tumor area （P 〈0. 017）. In the area of cirrhosis,the expression was significant lower than that in tumor area （P 〈 0. 017 ）. No significant difference was seen between cirrhosis and normal liver （P 〉 0. 017 ）. The positive expression rate of P-gp was significantly correlated with Edmondson＇s histological grade （P 〈 0.05）. Conclusion It is possibility that overexpression of P-gp promote the development and malignancy generation of HCC.
